The Basics

What Is Retatrutide?

First of all, retatrutide is an investigational once-weekly injection developed by Eli Lilly. It is often called a “triple agonist” because it acts on three hormone receptors: GLP-1, GIP and glucagon.

So far, it is being studied in large phase 3 clinical trials for adults with obesity or overweight and weight-related health conditions. Until the FDA reviews and approves it, retatrutide is not a prescription medication that clinics or pharmacies can legally provide.

The Three Pathways Retatrutide Targets

GLP-1

The same pathway targeted by semaglutide. It also helps with appetite, fullness and blood sugar.

GIP

Also targeted by tirzepatide. In addition, it is involved in insulin release and metabolic balance.

Glucagon

The new part. In other words, researchers are studying its role in energy use and how the body uses stored fat.

What the Research Shows

Retatrutide Clinical Trial Results

In fact, early phase 3 results for retatrutide have drawn a lot of attention. Still, clinical trial averages are not a promise, and more data are expected as additional trials are completed.

What researchers reported in TRIUMPH-1:

  • Adults with obesity or overweight, as well as at least one weight-related condition, without diabetes
  • Once-weekly doses of 4 mg, 9 mg or 12 mg, which were then increased gradually
  • Side effects such as nausea, diarrhea, constipation and vomiting were the most common
  • About 11% at the 12 mg dose stopped treatment due to side effects, vs. about 5% on placebo

Is It Safe to Buy Retatrutide Online?

No. Because retatrutide is not approved, any product sold outside a clinical trial has not been reviewed for safety, quality or correct dosing. In fact, Eli Lilly has stated that retatrutide is legally available only to participants in its clinical trials.

Similarly, the FDA has warned consumers about unapproved GLP-1 products sold online, including products with the wrong ingredients or doses. Read the FDA’s warning.

Red flags to watch for:

  • Sold online as “research peptide” or “not for human use”
  • No prescription, so no medical evaluation is required
  • Shipped as a powder, so you mix it yourself
  • Sold by a seller, but not by a licensed U.S. pharmacy
  • Claims to be “FDA approved” retatrutide

Does Allure offer retatrutide in Riverside?

No. Allure does not prescribe retatrutide. We offer medically supervised programs with FDA-approved options, such as semaglutide and tirzepatide, for patients who qualify.

When will retatrutide be available?

No approval date has been set. Eli Lilly is completing its phase 3 trials, and the FDA must review the results before retatrutide could be prescribed. Timelines can change.

How is retatrutide different from semaglutide and tirzepatide?

Semaglutide targets GLP-1. Tirzepatide targets GLP-1 and GIP. Retatrutide targets GLP-1, GIP and glucagon, which is why it is called a triple agonist.

How much weight did people lose on retatrutide?

In the TRIUMPH-1 phase 3 trial, people on the highest dose lost about 28% of their body weight on average after 80 weeks, compared with about 2% on placebo, according to Eli Lilly. Results vary and have not been reviewed by the FDA.

In trials, the most common side effects were digestive, including nausea, diarrhea, constipation and vomiting. They were more common at higher doses.
